At 3.15pm the man veered his falcon utility vehicle, towing a trailer, off the road and into a paddock.
There were no passengers in the vehicle.
Ballarat highway patrol senior constable Travis Johnson said alcohol and speed were not a factor in the crash.
"We are urging any witnesses, that we haven't spoken to, to contact Crime Stoppers or Ballarat Highway Patrol."
Ambulance Victoria spokesman John Mullens said it was believed the car had run off the road and come to rest in a nearby paddock.
Mr Mullens said preliminary indications showed there was no trauma involved and the man believed to be in his sixties may have suffered a cardiac arrest.
Police, ambulance and Country Fire Authority teams were called to the accident around 3.30pm and the road was blocked for over an hour.
Police are continuing their investigations.
A report for the coroner is being compiled.
