The crash occurred near roadworks at the intersection of Andersons Road and Western Highway, near Buangor, close to 3pm.
A female in her 20s was transported to Ballarat Base Hospital with ankle and rib injuries in a stable condition.
Another female, also in her 20s, was transported to hospital in Ararat for observation.
Ararat Police Sergeant Matt Horan said one woman was removed from her car with assistance from the SES.
Paramedics, fire crews, police and SES attended the scene.
