Police were called to Levuka Street, Seaford, after reports of gunshots about 9pm on Saturday.
A man aged in his 50s suffering from gunshot wounds was taken to The Alfred Hospital in a stable condition, an Ambulance Victoria spokesman said.
A woman, also with gunshot wounds, was taken to Frankston Hospital in a stable condition.
Victoria Police spokeswoman Creina O’Grady said a 29-year-old man was in custody and was being interviewed over the shooting.
She said the shooting was believed to have been connected to a neighbourhood dispute earlier on Saturday night.
