Police believe the woman was driving north along Bloomfield Road when she lost control on a sweeping bend and crashed into the tree between Old Sale Road and Telegraph Road around 3.40pm
The female driver, who is yet to be formally identified, was the sole occupant and died at the scene.
Traffic is being diverted and police will prepare a report for the Coroner, with investigators appealing for witnesses to come forward.
Police are also asking for help in investigating another single-vehicle crash, which occurred in Melbourne.
Last Tuesday afternoon it is believed a blue Mitsubishi sedan crashed into a message sign on the Mornington Peninsula Freeway, between Thompson Road and Peninsula Link just after 4.15pm.
The car's sole occupant, a 22-year-old Carrum Downs man, was taken to The Alfred hospital with life-threatening injuries.
Police do not believe any other vehicles were involved in the incident.
Anyone with any information on either incident is urged to contact Crime Stoppers by submitting a confidential crime report at www.crimestoppersvic.com.au or by calling 1800 333 000.
Victoria's road toll now stands at 194, 19 more than last year.
