Lumen Christi paired with Delacombe Primary School for the Music: Count Us In initiative.
About 600 pupils from the two schools sang simultaneously with 500,000 others nationwide at 12.30pm.
The Music Council of Australia initiative promotes the need for music education in schools.
Lumen Christi choir teacher Leonie Leviston said this was the second year the Delacombe primary schools had united for the event.
"We have done this for a few years, but for the last couple of years we have joined forces," Ms Leviston said.
Choirs from Lumen Christi and Delacombe sang Paint You a Song at Lumen Christi's activity centre.
Lumen Christi teachers helped pupils learn the song by playing it over the intercom at lunchtime.
Ms Leviston said it was important for school communities to celebrate music.
"We wanted to raise awareness of music and the joy it can bring to people's lives," she said.
"We all know the benefits that music can have for literacy, confidence and self expression."
Ms Leviston said a number of Ballarat schools also took part in the initiative.
"The kids look forward to it," she said.
Lumen Christi already has a strong focus on music education.
"The school already has a performing arts subject and all classes are involved in that," Ms Leviston said.
"It involves music and drama. Kids can also learn recorder, ukulele, xylophone and kazoo."
