About 80 Metropolitan Fire Brigade members were called to the building at the corner of Liardet and Bay streets at about 7am.
The two-story brick building houses a Telstra shop, a massage parlour and a cafe, and the fire caused about $2 million damage, a MFB spokesman said.
Two men, one aged in his 80s, were taken to The Alfred Hospital to be treated for smoke inhalation, an Ambulance Victoria spokesman said.
Both were in a stable condition.
Wind blew smoke in an easterly direction from the fire and the MFB advised locals to shut their windows and turn of air-conditioning units.
Twenty fire trucks were at the scene and brought it under control in about an hour.
He said a number of local residents were evacuated from their homes as a precaution.
The fire is believed to have caused about $2 million damage.
The MFB will investigate what caused the fire.
