This will be his first start since late 2012 – shortly before he suffered foot problems.
Wiltshire, 24, said it was great to finally be back in top shape and back on the track after having such a bad run with injuries.
He said he had put in a full pre-season and was in the best condition since he won the 2012 Stawell Gift.
As well as the 120m gift in which he is off 4.25m, he will line up in the 70m on 3m at Warrnambool.
Wiltshire said his aim was to make two finals, with his best chance in the 70m.
He will not back up in Sunday’s Terang Gift owing to work commitments.
