As part of an investigation into liquor licensing, police executed a search warrant at a Bourke Street venue about 2am on Sunday.
A 55-year-old man is assisting police with their inquiries.
A Victoria Police spokeswoman said a number of items were seized during the raid, including a significant quantity of alcohol.
It is the third time police have been to the venue this year.
Senior Sergeant Dave Sheppard, from the specialist taskforce "Razon" that monitors licensed venues across the state, said it was hoped the early morning raid would help reduce alcohol-related harm in the city.
He told Nine News the venue constantly trades past their licence time.
"They have a licence time of 1am. When we came here today, which was at 2am, they were still trading, serving liquor to their customers and allowing them to consume liquor on the premises," he said.
"We need to ensure that venues continue to be held accountable for any breaches in their licence.
"The things we look for at these venues are issues such as trading outside of their licence times, serving alcohol to an intoxicated person, permitting intoxicated people onto licensed venues and poor security.
"We want to reduce any possible harm to the licensees and the patrons who attend these venues," Senior Sergeant Sheppard said.
