Herbert ended Saturday on crutches as the Roosters went down to Essendon by 38 points.
Coach Marc Greig said Herbert “heard a pop” – which is often a tell-tale sign of a player who has ruptured their Achilles tendon.
However, the Roosters remain hopeful it is only a strain and will know more in the early stages of the week.
Meanwhile, Matt Johnston was left nursing some sore ribs after a heavy hit.
The Roosters were unsure on the extent of the injury, but x-rays would reveal if there were any cracks.
“You know when he goes down he obviously got hit pretty hard,” Greig said of Johnston.
Brenton Payne left the ground in the first quarter with a cork while Matt Austin was a late withdrawal due to a shoulder complaint.
Greig said the club would be taking no risks on injured players.
