Holman, a past Australian PGA Championship winner, notified tournament organisers on Friday that he would not be attending this weekend’s competition due to illness.
Holman claimed the 2015 PGA title in a play-off victory over the United States’ Harold Varner III and South African Dylan Frittelli.
Despite his absence, the new two-day event in Creswick has a strong field.
Among those is Ashley Hall, who was second in the 2016 Australian Open; US Champions Tour member and past Victorian PGA Championship victor David McKenzie; and Matthew Millar, who won the NZ Masters last week.
Andrew Martin, Josh Younger and Jack Wilson, who all secured 2018 Asian Tour cards last week, are also on their way.
Saturday’s round, which tees off from 12.30pm, will see professionals compete over 18 holes of stroke play and amateurs battle it out over 18 holes of Stableford.
The top 24 players in each category then progress to a series of six-hole match play elimination games on Sunday to decide overall winners.
The competition is based on next month’s ISPS HANDA World Super 6 Perth.
RACV Goldfields Resort’s last major golf tournament was the 2013 Victorian PGA Championship.
