Knowles belted the Roosters bowlers to all parts of Northern Oval 2 in a brilliant knock of 144, guiding the visitors to 344 before being bowled out late in the afternoon.
Nathan Edwards (75) and Tim Knowles (43) were the other major contributors for the Bulls.
North reached stumps at 0/18 and has a big task ahead next week if it is to reel in the massive first innings total.
