The Highlanders scored 2-124 in reply to the Knights' 7-101 in the 40 over clash at Coburg.
The victory follows triumphs over East Metro and North Metro in Twenty20 matches on Tuesday.
Coach Nicole Martin said the girls played exceptionally well, easily overhauling the Knights' total.
"The field was slow, which was our biggest challenge," Martin said.
"They just had to stay patient and push for singles. They were quite exhausted at the end of it, there were only three boundaries so they just ran the entire time."
Sarah Tatchell made 60 and Jenna Bruton made 50 not out, both batting for the 40 overs.
Highlighting the fine fielding performance, the Highlanders effected three run-outs, their first for the tournament.
They will now meet Northern Rivers at Plenty Valley for another 40-over clash tomorrow.
"Hopefully we'll be playing some more good cricket tomorrow," Martin said.
Central Highlands are currently in equal second in the competition, on 12 points.
"If we win tomorrow, we should be through to the finals at the MCG on February 24," she said.
