A police media spokeswoman said it was believed the prime mover was travelling west when it lost control and veered off the freeway onto an embankment, before rolling.
The driver, 47, was pinned by the legs in his cabin for several hours.
Paramedics administered pain relief to the driver until a crane was able to lift the cabin off the man a short time ago.
Ambulance Victoria spokesman John Mullen said the man suffered leg and pelvic injuries and was airlifted in a serious but stable condition.
"The driver was trapped for more than three hours," Mr Mullen said.
"They had to bring a crane in to lift the cabin to take pressure off his legs."
VicRoads blocked traffic on the freeway in both directions, from the Ballarat-Daylesford Road exit at Brown Hill, to the Victoria Street exit at Woodmans Hill.
All lanes have been reopened.
