The rider, a 55-year-old man from Sebastopol, collided with a Ford station wagon at the intersection of White Road while travelling towards Ballarat.
"All we can say is there's a collision with a motorcyclist versus a car, and we're investigating the cause of the collision," said Acting Sergeant Wayne Jones.
The rider died at the scene.
Leading Senior Constable Lee Thomson from Victoria Police's media unit said the 27-year-old driver of the Ford was arrested at the scene.
However, the man has since been released pending further inquires.
"It’s believed the station wagon was travelling north on Whites Road before entering the intersection with Cuthberts Road," he said.
Ballarat Criminal Investigation Unit detectives are investigating the crash, and police have asked anyone who saw either vehicle to come forward.
