Ms Harris is undergoing treatment for breast cancer and Ms Cooke has just been diagnosed with it.
“I’ve got two more rounds of chemo to go and then radiation,” said Ms Harris.
“I’ve just got a job and everything’s got positive.”
Ms Cooke said she drew strength from people who had survived cancer.
“To see all the survivors here, you think ‘they’ve done it, so I should get through it’,” Ms Cooke said.
The Llanberris oval event raised more than $80,000 for cancer research.
Coordinator Rob Petrie said the community strongly supported the gruelling all-night walk around the track.
“We’ve had more volunteers this year than ever before,” he said.
“We’ve had more donors and sponsors than ever before.”
Relay for Life Ballarat chairman Andrew Kilmartin said on Sunday there were 743 participants and $84,000 raised.
