Zoe Denahy, Pip Metcalfe and Ella Friend will line up against Calder Cannons at Mars Stadium on Saturday.
Denahy and Metcalfe hail from Lake Wendouree to add to an already big presence for the Lakers and Friend is from Horsham.
This adds to six players who made their debuts in the opening round.
Rebels won their round one assignment - defeating Bendigo Pioneers by five points on March 2.
The whole competition had a week off ahead of this weekend's fixture.
The Rebels' first appearance at home will also feature a futures game, allowing the whole squad to be in action.
This will be a curtain-raiser at 11am, with the NAB Cup match at 12.45pm.

THE Rebels boys' squad continues its NAB Cup under-18 pre-season with practice matches against Geelong Falcons at Mars Stadium on Sunday.
The under-16 development squad plays at 11am followed by under-18 matches at 1pm and 3pm.
GWV Rebels begin their home and away season against Dandenong Stingrays at Mars Stadium on Saturday, March 23.
