Premiership wing defence Lauren Gilcrist will play out the milestone game in her 22nd season with the Lions.
Gilcrist has helped lead the Lions to multiple premierships since 1997, including their 2017 grand final victory over Lake Wendouree.
The 38 year old laughed when she spoke about the milestone, saying all it did was "show her age."
Gilcrist met her husband through the club and now has two children playing sport with the club.
"I grew up in Sunbury and have stayed with the club because of the amazing friendships and people I've met," she said.
Gilcrist's family and friends and set to attend the game to celebrate the milestone with her.
She would not confirm whether she would return to the Lions for a 23rd season, saying her children were now her priority.
