To be screened on ABC1 later this year, the drama stars Craig McLachlan and Nadine Garner and will begin filming later this month.
Ballarat Regional Tourism director George Sossi said traders along Lydiard Street had been notified about road closures during filming.
Filming is believed to be scheduled to start on April 16.
“We’ve already had preliminary discussions with traders along Lydiard Street, but there will be more (discussions) once we’ve seen the official schedule,” Mr Sossi said.
The series revolves around Dr Lucien Blake, who returns home to Ballarat in 1959 to take over his late father’s medical practice.
But Dr Blake (McLachlan) is also a “keeper of secrets and a solver of mysteries”.
The series is written by John Alsop and will be produced by Tony Wright and George Adams of December Media, which mainly produces documentaries, live-action dramas and animated children’s television shows.
December Media is particularly renowned for its large-scale historical documentaries, including the four-part $4.5 million series Captain Cook: Obsession and Discovery, Revealing Gallipoli and the popular factual series Family Footsteps.
December Media has said The Doctor Blake Mysteries would be produced in partnership with UK-based ITV Global and ABC Australia.
The series will be filmed in Ballarat and Melbourne between April and August.
