Daylesford kicked two goals just before the 80th minute to snatch the game 3-2 at Victoria Park on Sunday.
The win locks-up the league title for the club, and puts a stamp on what has been a Cinderella-like return to the premier competition.
Daylesford now enters the post-season with a double chance, and one win away from a grand final berth.
The result could be potentially dire for Warrnambool however, which has dropped to third following the loss, and now faces a do-or-die encounter with Ballarat North United in the first week of finals to keep its season alive.
Elsewhere the Sebstopol Vikings locked-up second spot after thrashing Maryborough 13-0 at home.
Ballarat North United defeated Victoria Park 3-2, Bacchus Marsh was easy winners over Ballarat, while Forest Rangers thumped Creswick.
Meanwhile Ballarat will enter finals in top spot after defeating Ballarat North United 2-0 in the division one women's competition.
The club will hope the result is a sign of things to come when it takes on Ballarat North again in the first week of finals this weekend.
In the other side of the pool, the Forest Rangers will take on Victoria Park. The match-up will be unlikely to phase the Rangers following their 5-0 thrashing of Victoria Park in the final week of round play on Sunday.
