The incident occurred about 12.40pm on Wallis Street, at the corner of Martin Drive, in an industrial estate.
While neither driver was injured, two people were assessed at the scene, with one, believed to be a passenger, was taken to Ballarat Base Hospital, an Ambulance Victoria spokesperson confirmed.
The full extent of the damage to the rare Australian vintage automobile was unknown but it had visibly suffered significant panel damage to its front end and grill.
The iconic model of early Australian motoring, often restored meticulously as a labour of love, was taken away by a tow truck.
The Holden 48/215 was produced between 1948 and 1953.
