Investigators believe a collision involving up to three cars occurred on Bacchus Marsh-Geelong Road in Balliang, at the intersection of Birds Road and Sharkey Road, just after 1pm.
A man and a woman from separate vehicles died at the scene.
A boy has been airlifted to hospital with life-threatening injuries.
Another man was taken to Geelong hospital for non-life-threatening injuries.
Major Collision Investigation Unit is enroute to the scene.
The exact circumstances surrounding the collision are yet to be determined and the investigation is ongoing.
