She added to her 2012 and 2016 titles - also Olympic years - with an aggressive ride in the FedUni Cycling Australia Road National Championships in Buninyong on Sunday.
The gold medal further consolidates Spratt's status as one of Australia's all-time great cyclists.
The performance also reinforced her longevity at the top as the 32-year-old heads to another Olympic Games in Japan later this year.
Spratt said the road race was her priority for the Olympics, where she was confident Australia could secure gold.
The New South Welshwoman led Justine Barrow and Mitchelton Scott teammate and past national time trial champion Grace Brown over the line for the podium places.
The three formed a breakaway in the third of nine laps and were never challenged.
Spratt said the Mitchelton Scott team plan had been to break up the field early, but she was surprised to have just three in the breakaway - not only initally, but for the balance of the 104.4km event.
Brown was dropped in the closing 30km, while Barrow had no answer to Spratt over the last 500m.
Spratt said she was always felt confident that she could out-sprint Barrow.
Spratt said he was grateful that the team had gone with the plan, and particularly grateful for the support of Brown, who finished 10 seconds behind Spratt and Barrow.
"This is special. I really really wanted it."
She said there was a lot of pressure pre-race and she had consequently felt a little more nervous than usual.
Spratt now chases a fourth consecutive Women's Tour Town Under in South Australia.
