The man was killed after smashing into a tree on Ellis Road in Coomoora about 1.30am.
Emergency services were called but he was unable to be revived and died at the scene.
Coomoora is about five kilometres north-east from Daylesford.
The investigation into the exact cause of the collision remains ongoing.
The man is yet to be formally identified.
Ellis Road is a narrow, unsealed back road in Coomoora, lined with a number of houses and semi-rural properties.
The death on an unsealed road follows a similar incident near Clunes last week.
In that fatality, a driver lost control and crashed through a fence along Clunes-Mount Cameron Road, with the car bursting into flames.
The latest fatality brings the road toll to 15 for the year in Victoria, six more than at the same time last year.
