There has been no new cases confirmed in the state since yesterday.
The last day Victoria had zero new cases over a 24-hour period was on March 5, before the state of emergency was put in place.
"We've been able to achieve this through Victorians doing an incredible job keeping themselves and each other safe," Ms Mikakos said.
"The public health advice is that the risk is not over yet so it's up to all of us to keep it up.
"Let's keep up this tremendous effort."
Victoria's total number of recorded COVID-19 cases remains at 1,681. Of those, 1,586 were declared as recovered on Friday.
This weekend is the first long-weekend since the easing of lockdown restrictions has allowed travel and overnight stays.
Black Lives Matter rallies are set to be held across Australia today, including in Ballarat, to show solidarity with the movement and African American George Floyd who died while being arrested in Minneapolis.
The Australian protests are also a show of support to the Aboriginal community to highlight high levels of indigenous incarceration and deaths in custody.
Prime Minister Scott Morrison has urged Australians not to attend the protests.
Victoria Police has warned rally organisers could be fined $1651 because of COVID-19 restrictions and that fines could also be issued to individuals.
Premier Daniel Andrews told reporters on Friday protesters should find a safer way to make their point.
"We don't want all the hard work, all the sacrifice that Victorians have made, frittered away by people gathering to make an important point, but it is not the only way to make that point," he said.
