Glover from Dimboola has been off the scene for two years after a knee reconstruction.
She won the Rebels' under-16 rookie award in 2018.
Glover was set to play her comeback game in round two, but the NAB League under-19 completion was postponed owing to the Victorian COVID-19 lockdown.
As well as welcoming back Glover, the Rebels will debut Olivia Moll from Ballarat Grammar School.
She has been named on interchange.
Head coach David Loader said Glover had done a power of work, and been a great role model and leader within the program.
Loader said challenging times would show the character within the group.
"Things are easy when all is well, but the character will show out when things become a challenge."
The Rebels lost to Geelong in round one.
