It is understood the male was travelling south on Navigators Road, just past Ramsays Road, just after 9am when he left the road.
His Toyota SUV collided with a tree and rolled.
The only occupant, he was ejected from his vehicle and sadly died at the scene.
Moorabool Highway Patrol Sergeant, Jarrod Chatfield, said the male driver was believed to be local to the area.
Police are still in the early stages of their investigation, which will include assessing a range of potential causal factors.
"We are still in the early stages of the investigation but we will look at the overall picture of speed, alcohol, drugs and everything else we look at in every collision," Sergeant Chatfield said.
Sergeant Chatfield said it was not believed any other vehicles were involved.
Police have cordoned off the area as the scene is processed and investigated.
The section of road will be closed for several hours.
Police are in the process of informing the man's family.
Sergeant Chatfield encouraged motorists to drive to the conditions.
"Conditions change so driving style should too," he explained, adding changes to road surfaces, such as roads being after rain, should determine changes to speed.
Moorabool Highway Patrol is in charge of the investigation.
