The Ballarat product received the peer-voted honour in recognition of his influence on the playing group and the selfless work he's done with the club's community programs since joining from the Western Bulldogs in 2019.
Roughead also finished eighth in the best-and-fairest count after a solid season in defence.
"It's a real honour to be awarded the Darren Millane Trophy," Roughead said.
"I'd like to thank everyone who voted on the award, my teammates and the rest of the football department and coaching staff.
"It would be remiss of me not to mention my wife Bridget. Her support allows me to be the person I am around the football club. So, big thanks to her.
"Also to Mum and Dad, big shout out to you."
The 30-year-old was nominated for the AFL's Jim Stynes Community Leadership Award for a second time this year.
Among the organisations Roughead has worked for include being an ambassador for not-for-profit association Stand-Up Events which works to help educate young people on the impacts of homophobia.
He has also participated in a world-first research project with Monash University into homophobia in sports clubs.
Roughead is also active in generating awareness of climate change through his involvement in the environmental organisation FrontRunners.
Roughead has also been a long-time supporter, ambassador, mentor and board member with Ladder, working to support young people who experience homelessness and social isolation and disadvantage.
He is also an assistant coach for Collingwood's AFLW side.
