The driver, 21, from Ballarat, was driving a Ford sedan on the Western Highway heading towards Ballarat with another male passenger,18, when he lost control of the car near the corner of Black Bottom Road in Brewster, just before 1pm on Saturday.
Learmonth Leading Senior Constable David Kelly said the car veered onto the other side of the road before hitting the embankment, flipping in the air and rolling several times before landing upward on the side of the road.
He said four police vehicles, three ambulances and two CFA units attended the scene.
"The CFA rescue unit cut the roof of the car off to extract the driver who was trapped inside," Leading Senior Constable Kelly said.
"A helicopter arrived shortly after and he was then immediately flown to Melbourne with back, chest and arm injuries,"
He said the passenger from Bairnsdale sustained minor cuts and grazes and was taken by ambulance to Ballarat Base Hospital for observation.
The driver remains in a stable condition at the Royal Melbourne Hospital.
No other vehicles were involved in the incident.
Leading Senior Constable Kelly said police wanted to speak the driver of a car that is believed to have witnessed the incident.
He said police are still determining the cause of the incident and investigating whether speed was a factor in the crash.
He urged anyone with information to call Crime Stoppers 1800 333 000.
