The woman, aged in her early 60s was the sole occupant of the silver Holden sedan which left the road about two kilometres north-east of Stawell just before 7.30am.
Stawell Police Sergeant Bill Alford said the female driver, who died at the scene, appeared to have lost control of the vehicle and hit a tree.
"There is no indication fatigue, alcohol or drugs played a part in this crash," he said.
Police and emergency services responded to the incident. Stawell-Navarre Road was closed to traffic for a couple of hours as investigations commenced.
Sgt Alford said a report would be prepared for the Coroner.
The single vehicle fatality takes Victoria's provisional road toll for 2014 to 199, 16 more than for the same period to date (183) in 2013.
