More than 5000 people dressed in black clothes are expected to attend Sunday's rally outside the State Library of Victoria from noon.
Organisers say the rally would be a display of solidarity with the US Black Lives Matter campaign, but would also highlight Indigenous Australian deaths in custody that are "taking place on our own soil".
"Stand in solidarity to put an end to police brutality, racial profiling, Indigenous Australian deaths in custody and injustice towards people of colour," the rally's social media page says.
"Please bring a peaceful mindset. No violence and no weapons."
The rally comes after the shooting death of two US men, Alton Sterling and Philando Castile, at the hands of police last week.
Five police officers were then shot and killed at a Black Lives Matter protest in Dallas, Texas, on Friday.
