The crash occured at the intersection of Clyde Hill Road and the Midland Highway about 4.30pm.
It’s believed a Nissan sedan, travelling southwest on Clyde Hill Road, collided with a 4x4 utility, towing a caravan, on the Midland Highway.
The driver of the sedan, a 79-year-old man, died as a result of the accident while his passenger, a woman in her 70s, was airlifted to a Melbourne hospital.
The occupants of the utility, a man, a woman and a small child, were conveyed to a Geelong hospital with minor injuries.
A police investigation is ongoing.
Seventy people have died on Victoria’s roads already this year.
