Holman, who won the national crown in 2015, knows the Creswick course well.
He was an early leader in the Victorian PGA Championship there in 2013.
The event has attracted more than 50 professionals including Chris Wood, Matthew Millar, Andrew Martin, Mitch A Brown and Mitch Brown, who all finished in the top 10 in national pro-am order of merit.
The experienced Scott Laycock and David Bransdon are also set to play.
The PGA-sanctioned event is part PGA pro-am series, making it also also open to amateurs.
Day one is an 18-hole stroke event for professionals and Stableford for amateurs, with the top 24 in each category making the cut.
Day two comprises six-hole match-play knockout series to find the winners.
