The Bulls went through Friday's middle session without losing a wicket to reach 2-185, for a lead of 26 runs at North Dalton Park.
Queensland suffered a blow before play with confirmation of a concussion, suffered by Matthew Renshaw on Thursday in training, leaving the in-form opener in doubt for the Shield final at Allan Border Oval.
The situation on the field was better for Queensland, however, with NSW adding just 20 to their overnight 7-382 to be all out for 402.
In reply, the Bulls stumbled early as Trent Copeland (2-26) struck twice to leave the visitors at 2-53.
Labuschagne (79no) batted confidently throughout the afternoon and, with Hemphrey (37no), they have added 132 for the third wicket.
