Spence, who is a member of the Ballarat Harriers, finished as the third Australian across the line and 32nd athlete overall in the annual distance event in Germany.
His time of 2:18:43hr is quick enough to be picked for the 2018 Games, which will be held on the Gold Coast during April next year.
Spence finished behind Kenyan Eliud Kipchoge, who won the race in 2:03:32hr.
He was placed behind fellow Australians Liam Adams (2:12.52hr) and David Criniti (2:17.57hr).
Spence, who is originally from Anglesea, won his third consecutive 44km men’s Great Ocean Road Marathon in May.
Legendary Ballarat distance runner Steve Moneghetti was named as one of the Berlin Marathon Hall of Fame inaugural inductees this year.
The honour came close to three decades after Moneghetti raced to victory in the 1990 event.
