The man believed to be about forty years old lost control of his car while driving east on the highway at about 7.30pm on Sunday and slammed into a tree near Mac’s Lane.
A police spokesperson said the single occupant suffered serious but non life-threatening injuries.
Shortly after 9.30pm emergency services manged to free the man, where he was treated on scene in preparation for him being flown by air ambulance to a hospital in Melbourne.
The Midland Highway remains closed at Buninyong west of the township near the Buninyong Golf Course.
Drivers are urged to find an alternative route.
