The Lakers managed to keep Darley scoreless at Darley Park on Sunday, while extending the margin every quarter to lead by 75 points when the final siren sounded.
While the Lakers are new to the competition, Darley has also introduced its first open age side this season.
Both clubs were looking for their first victory heading into the game after taking time to adjust to senior level.
Lake Wendouree coach Melinda Sands said it had been a special moment to head into the rooms and sing the song for the first time.
“It was a very exciting game for us, obviously, getting our first win for the season,” she said.
“We were strong for four quarters and all of the hard work and learning and commitment that we’ve been putting into place right from preseason through to now came together.”
Lakers’ Lauren Mclean booted four goals, while Jacqui Clifton had three.
“Lauren Mclean was best on ground, but we had quite a few contributors to the goals and we had some really good strength off centre half-back from Ella Bibby, which was her first game with us,” Sands said.
“It was a free-flowing game from us.”
Meanwhile, Redan enjoyed an emphatic 67-point victory over Melton Centrals as it continues its run as the only undefeated team in the competition.
East Point got on top of Gisborne in the second half of its away game to win 20-49.
While Carisbrook enjoyed the biggest margin of the round seven clashes with a 85-point on-the-road victory over Ballarat.
