Learmonth coach Jordan O'Keefe said the calf injury came as a surprise to O'Brien who has been fairly durable over her sporting career.
"She's never really been injured before so it gave her a bit of a shock," he said.
"We just thought we would give her a few weeks to recover and time it so she can return after the Queens Birthday bye."
The Lakies will miss two other defenders ahead of the clash with the Saints, leading O'keefe to say the contest will be a good test of their versatility.
