It is understood a woman is trapped in her car after the crash near Cardigan, which occurred about 1.30pm.
Police, CFA rescue crews and Ambulance Victoria are on scene of the two-car crash.
Remembrance Drive is blocked in both directions near Gleno Court.
Rescue crews are working to remove the side door of the car, in order for paramedics to treat the patient.
There are five CFA units on scene.
One person has suffered upper body and leg injuries and has been airlifted by helicopter to the Royal Melbourne hospital in a serious but stable condition, according to an Ambulance Victoria spokesperson.
People in the area are advised to exercise caution.
