Last year's premier scored 12 runs in the final two overs of the second innings to win the match at Russell Square Oval on Saturday.
East captain Joshua Brown's timely four brought the victors within a single run of the target in the second last over, allowing his team to take the match with five balls remaining.
East Ballarat opening batsman Lewis Hodgins said it was good to record the first win of the season.
"We wanted to keep them under 250 at the start of the day, so to get them at 213 we were pretty happy," he said.
"With one run to win in the last over they brought their field right up, so we were pretty confident of hitting over the top and we did."
Chris Jerram's knock of 59 laid the platform for victory for East Ballarat, while Brown finished on 30.
"We are still waiting for our top-order to strike and when that happens we'll be a very good all-around team," Hodgins said.
Meanwhile Golden Point's Josh Pegg recorded a century to lead his side to a 74-run win over Buninyong.
The opener scored 101 off a 128 balls in the match played at Buninyong. He was backed up by Josh White with 60 and Daniel McDonald who took three wickets.
Elsewhere Nap-Sebas failed to run down Wendouree, Darley enjoyed a 60-run win over Mt Clear and North Ballarat beat Brown Hill.
