Emergency services were called to the crash on the corner of Clayton and Joseph streets about 2.50pm.
One person was taken to Ballarat Base Hospital, Ambulance Victoria confirmed, but the other driver was unhurt.
WHERE THE CRASH HAPPENED
Police at the scene said it's alleged the truck driver failed to give way while driving south on Joseph Street - the car travelling west along Clayton Street attempted to brake but could not stop in time.
The truck then tipped on its side.
Witnesses said it "all happened very fast", and the truck appeared "unbalanced" as it went through the intersection.

"I heard a bloody loud bang," another resident said, noting this was not the first accident they had seen on this stretch of road.
"It's a rotten intersection, it's a wonder (the truck driver) didn't get killed."
A heavy duty tow truck was called in to right the truck once the debris was cleared.
