A spokeswoman for Ambulance Victoria said paramedics were called at about 2.10pm to the incident at Vite Vite.
"One person is being airlifted to the Royal Melbourne Hospital and two others are being taken by road to Ballarat Base Hospital," she said.
"There are no details about injuries, conditions or genders at this stage."
The Courier understands the accident involved a freight train and a truck at the railway intersection of Vite Vite Road and Mercers Road.
Police sources said emergency services and heavy haulage were on the way to the scene.
