Police have confirmed the incident this morning was a single-car crash into a tree.
The driver and sole occupant of the car, a 32-year-old Ballarat man, died at the scene.
Police told The Courier it was believed the man was travelling towards Beaufort when he veered on to the opposite side of the road and crashed into a tree.
The road reopened about an hour ago.
Multiple police and firefighter units were at the scene for several hours, with part of the road blocked from both directions as it was processed by investigators.
The section of road reopened to traffic just before midday.
Police are continuing to investigate.
It is the twelfth fatality on roads across the Ballarat region in the last 12 months. There have been 220 lives lost on the road this year, compared to 195 at the same time last year.

A man has been killed after crashing into a tree near Beaufort on Friday morning.
The crash occurred about 7.10am on Beaufort-Carngham Road, about two kilometres south of Beaufort.
The man died at the scene.
Police confirmed the incident to The Courier this morning, stating the man was the sole driver of the car.
Investigators are yet to establish the circumstances of the crash.
The road is heavily lined with trees on the side of the road where the crash occurred.
Multiple police and firefighter units remain on scene and the road is expected to be blocked for some time.
The fatality was the second for the morning in Victoria, with another life lost in Springvale South.
Emergency services were called to Springvale Road about 5.30am after reports a car had crashed into a pole.
The male driver and his female passenger were freed from the vehicle and taken to hospital in critical condition.
