Victoria Police have confirmed they attended the massive queue at the Creswick Road testing site, amid safety concerns.
According to Victoria Police Media, police responded to reports of "significant traffic issues".
"The traffic congestion caused safety concerns, police consulted with site management at the location and the site was suspended by the provider," a spokesperson said in a statement.
They referred further enquiries to the state Department of Health.
The testing site, the only drive-through clinic in Ballarat, will be closed all of Wednesday, with Ballarat Health Services tweeting it will continue to close in periods of high demand "for the safety of our staff and those attending".
Ballarat Health Services has been contacted for further comment.

Ballarat Health Services' COVID-19 testing site in Creswick Road remains closed without any indication of when screening will resume.
Motorists seeking tests continue to be turned away from the drive-through site this morning.
The Courier understands some people had been queuing in early hours this morning in the hope of being tested after the site was unexpectedly shut on Tuesday, due to overwhelming demand.
BHS has issued a media statement saying testing will remain suspended to clear a backlog of testing from the site.
The statement also read the site would continue to shut down in times of high demand to protect the safety of staff and attendees.
The 4Cyte Pathology drive-through testing site in Redan has remained closed since Christmas.
UFS' COVID-19 clinic has continued testing about 200 to 300 people each day but, due to limited resources tests are by booking only.
If you have a positive result from a rapid antigen test, you must consider yourself COVID-positive, isolate at home and get a PCR test (polymerase chain reaction test) from a designated site, such as Creswick Road or UFS.
Testing via Daylesford Health is also booked out for today, Central Highlands Rural Health has confirmed.
EARLIER, Tuesday, January 4:
Ballarat Health Services' Creswick Road COVID testing site remains closed, as a backlog of tests already taken is cleared.
In a post to its Facebook page, BHS said the clinic is not taking any further vehicles until the backlog is cleared and a new triage system is implemented.
"The Creswick Road testing site is only testing people if they: are household contacts of an already confirmed positive case, or are symptomatic,"
"Until further notice, if you test positive with a RAT, you are considered COVID positive. Please isolate at home."
Anyone having difficulty accessing a PCR test at the clinic today is asked to isolate a home and either book a test with UFS or return to the Creswick Road testing site tomorrow.
Anyone wanting to obtain a test, is urged to check back with BHS this afternoon for any further updates.
The 4Cyte Pathology drive through testing clinic in Redan is also closed, meaning there is no drive through testing capacity anywhere in Ballarat at this time.
The lack of testing, combined with a shortage of rapid antigen tests in Ballarat and across the state, has caused frustration and concern for those who have symptoms or are close contacts of positive COVID cases.
It comes as Victoria recorded 14,020 new COVID cases in the 24 hours to midnight, Monday.
